SUMMARY
This
thesis includes five chapters as follow :
Chapter
one :- This chapter consists of the first part, included an overview of flow injection
technique and its kinds and reference to some
of its applications , either the second part includes a general
review of amino
acids , uses , benefits and included some methods used in the estimation of amino acids
with different technologies , completed the
chapter by the objective of the search .
Chapter
Two :- Chapter two contains the chemicals and how to prepare them at different
concentrations used , in addition a detailed explanation
of apparatus and tools that were used in the study .
Chapter
three :- This chapter contains two parts. The first part includes a brief introduction about
arginine and its benefits for the body , uses ,
presence and its sources. The second part has included detailed presentation of the two
spectral methods , batch method and flow
injection / merging zones method to estimate arginine in
pharmaceutical and
vital samples using Sakaguchi reaction which was based on the oxidation of the amino acid
arginine by sodium hypobromite in acidic
medium
and then condenses arginine with alpha-naphthol reagent to form red colored complex measured
at a wavelength 501 nm . Also all
chemical and physical conditions affecting the reaction were investigated to reach its
optimal values over the study and it was 1-45
μg.ml-1 and 3-1400μg.ml -1 methods batch and flow
injection / merging
zones respectively, and detection limit for each method 0.139 μg.
ml -1 and
1 μg.ml -1 . Stiochemistry of the formed product was studied by using the molar ratio and
the job methods , the resulting found to be 1: 2
(reagent
: drug) . Stability
constant was studied the resulting found that the stability
constant
was 2.069x1010 (L 2. mol-2 ) or (M -2) . Batch and FIA /
merging zones were applied in the estimation of arginine in pharmaceutical preparation
and vital samples (grape juice ) , results was
compared statistically using t-test and F-test with the results of
the standard method
(European Pharmacopoeia) at a confidence level of 95%
as
it was found there is no significant difference in accuracy and credibility of the two methods .
Chapter four :- This chapter
contains two parts. The first part includes an introduction
about tryptophan and its benefits for the body , uses , presence and its
sources. The second part was included a detailed presentation of
the two spectral methods , batch method and flow injection /
merging zones method to estimate tryptophan in pharmaceutical
preparations using azo-coupling reaction which was
based on oxidation of the reagent
diphenyl sulphonate to diphenyl
benzidine sulphonic acid after
reaction with sodium nitrite in sulpharic acid as medium ,
the unstable oxidation product react with sodium nitrite to produce a
diazotized intermediate , which in turn coupled with tryptophan to
form a pink colored complex measured at a wavelength 522 nm .
Also, all chemical and physical conditions affecting in the reaction
were investigated to reach its optimal values over the study and it
was 3-40 μg.ml-1
and
5-100μg.ml -1
batch
and flow injection / merging zones
respectively, and detection limit for each methods 0.2325
μg. ml -1 and 1 μg.ml -1 . Stiochemistry
for the formed product was studied by using
the molar ratio and the job methods , the resulting found to be 1: 1
(reagent : drug) .Stability constant was studied and the resulting found that
the stability constant was to

FIA / merging zones
were applied for the estimation of tryptophan in pharmaceutical
preparations, results was compared to statistically using t test
and F with the results of the standard method (British Pharmacopoeia)
at a confidence level of 95% as it was found there is no
significant difference in
accuracy and credibility of the two methods .
Chapter five : -
Summarized
the conclusions which based on the result that obtained
from practical part which relates the use of new approaches for
the determination of amino acids ( arginine and tryptophan ) using the developed
FIA/ merging zones technique , recommendation and published papers
.
